Orihuela will face off with Villarreal in a Copa Del Rey match at the Estadio Municipal Los Arcos on Saturday.
Orihuela reached this stage after defeating Cornella 5-4 on penalties following 0-0 draw away in the first round tie last month.
They were eliminated from last season's competition by L'Hospitalet in the preliminary round.
Maxi Amondarain, the Orihuela defender and captain, will be looking to lead his side to a famous win over Villarreal.
Orihuela will be looking to stun a much-fancied Villarreal side which is looking to win this competition for the first time.
Meanwhile, Villarreal reached this stage after defeating Comillas 5-0 away in the first round tie last month.
The Yellow Submarine were eliminated in the Round of 16 by Espanyol in last season's competition.
Colombian centre forward Carlos Baccus is the dangerman for Villarreal having netted a hattrick against Comillas.

Javier Calleja, the Villarreal coach, has praised his charges ahead of their clash with Orihuela.
“We’ve started 2020 as we finished 2019. On the way up. The team knows what they’re doing and are giving the best of themselves," Calleja said.
"We’re very talented. We had to get stronger at the back and we’ve done just that.”
This will be the first competitive meeting between Orihuela and Villarreal.
